Induction of Apoptosis and Telomerase Activity of NCI-H460 Human Lung Carcinoma Cells by Methanol Extract of Houttuynia cordata Thunberg

Abstract
Houttuynia cordata Thunb. is a medicinal plant of the Saururaceae family widely used in folk medicine in several Asian countries. It has been reported that extracts of H. cordata exhibit anti-cancer activity through apoptosis induction, although the mechanisms are not fully understood yet. In the present study, it was investigated the further possible mechanisms of anti-proliferative effects by the methanol extract of H. cordata (MEHC) in cultured human lung carcinoma NCI-H460 cells. The results indicated that exposure of NCI-H460 cells to MEHC resulted in cell cycle arrest at G2/M phase and induction of apoptosis, which was associated with inhibition of the expression of cyclin B1 and activation of caspases, respectively. Moreover, MEHC inhibited telomerase activity through down-regulation of the expression of human telomerase reverse transcritase and c-myc. Taken together, together with previous findings, the data provide important new insights into the possible molecular mechanisms of the anti-cancer activity of H. cordata.
